Innovations In Perfusion Bioprocessing

Perfusion is a cell culture method becoming increasingly popular for producing monoclonal antibodies and their derivatives. It offers higher space-time yields (STY) and shorter residence time for recombinant proteins in the bioreactor, while also enabling various process intensification strategies. A successful perfusion process relies on media that supports rapid cell growth during the exponential phase as well as efficient protein production during the stationary phase, all while reducing media consumption. Transitioning from fed-batch to perfusion mode often involves developing a medium that can sustain higher cell densities and increased productivity without compromising product quality.

In this webinar, a method for deriving perfusion culture media by building on existing fed-batch media and feeds is demonstrated. This approach incorporates strategies for adjusting media depth to align with the cell-specific perfusion rate (CSPR) and addressing lactate overflow metabolism in perfusion conditions. Additionally, the successful scale-up of this perfusion process from small-scale experiments to a fully integrated continuous bioprocess at a 50 L pilot scale is showcased. This session provides valuable insight into optimizing and scaling perfusion media and processes, driving progress in the production of monoclonal antibodies in biotechnological applications.
