Developing A Large-Scale Tangential Flow Filtration Process

Tangential flow filtration (TFF) is a crucial tool in the downstream processing of biopharmaceuticals. In Cytiva's large-scale single-use TFF product line, a TFF cassette filter holder, connecting tubing manifold, and sample distribution plate have been added. This setup eliminates contact with multiuse surfaces both before and after filtration. The holder is compatible with membranes having at least 10 m² of surface area and supports the feed flow of the ÄKTA readyflux XL tangential flow filtration system.

A model concentration and diafiltration process of bovine γ-globulin was developed to showcase the combination of these single-use TFF solutions. Omega™ polyethersulfone (PES) membranes were employed for an initial sample volume of 500 L. This approach enabled a 10-fold volume reduction and a four-fold buffer exchange in under 2 hours using 5 m² of filter surface area.
