The Evolution and Future of Targeted Protein Degradation Chemistry

Targeted protein degradation (TPD) has long been one of drug discovery’s most compelling —but elusive—modalities. Now, with the recent FDA approval of Arvinas’ PROTAC therapy, vepdegestrant, the field is beginning to shift from theoretical potential to clinical reality.

“This approval has been a major step for the field,” said Maria Soloveychik, co-founder andCEO of SyntheX. “In terms of mechanism, it’s proof that bivalent compounds can work clinically.”

For many professionals in this space, the milestone signals a broader inflection point. After years of scientific groundwork, degraders are now being tested—and in some cases validated—where it matters most: in patients.

Johannes Yeh of Cold Spring Harbor Laboratory emphasized the growing confidence among researchers. “The clinical demonstration is definitely one of the biggest drivers,” he said. “There’s no question anymore that this modality can degrade proteins in vivo.” Yet, despite this progress, the field remains technically complex—and far from solved.
