Source: Sartorius
By Marcel Fueger, Yeonhwa Jin, Axel Thiefes, and Michal Szelwicki
Depth filtration plays an essential role in producing cell and gene therapies, particularly in clarifying adeno-associated virus (AAV) vectors. This study assessed the effectiveness of Sartoclear® depth filters in processing AAV8 vectors derived from HEK293 cell cultures.
Key Findings:
- Exceptional Performance: Small-scale experiments demonstrated high filtration capacity, excellent protection for downstream sterilizing filters, and strong product recovery rates.
- Scalability: Process-scale evaluations highlighted the robust and scalable performance of Sartoclear® DL75 filters, ensuring high recovery even with challenging feed materials.
Implications:
The use of Sartoclear® filters in suspension-based AAV processes enhances purification efficiency and product yield, representing a significant advancement in supporting the broader accessibility of cell and gene therapies for patients.
