Upstate Biotechnology is preparing the launch of its human G-Protein-Coupled Receptor (GPCR) product line. The GPCR membrane preparations will be available in test amounts for assay development and in bulk amounts for high throughput screening binding assays. Each membrane preparation is obtained from an adherent CHO-K1 cell line stably transfected with a recombinant GPCR. Membrane preparations that can be used for assay development are packaged at 200 assay points/vial. An assay point is defined as the amount of GPCR membrane protein that will confer >1000 CPM when binding its specific radioligand near the dissociation constant (Kd). Each lot of a GPCR membrane preparation is characterized by radioligand binding assays to determine its ligand affinity and receptor density. The physiological nature of the membrane preparation is confirmed by competition assays with agonists and antagonists. The GPCR membrane preparations are stored at -70oC and are stable for 6 months.
